A process has been developed for the separation of ten individual tRNAs from E. coli cells. This process consists of phenol extraction and two cycles of ethanol precipitation to produce a crude tRNA, followed by reversed-phase chromatography under a variety of column conditions to isolate the desired tRNA. For the processing scheme chosen, glutamic acid, two formylmethionine, arginine, valine, aspartic acid, lysine, normal methionine, and two phenylalanine tRNAs were isolated as high-purity products. Isoleucine, histidine, and alanine tRNAs were isolated as partially pure products. The quantitative tRNA recovery averaged about 10% per column run.
