A Two Generation Reproduction Study of P-nitrochlorobenzene in Rats with Cover Letter and Summary.

p-Nitrochlorobenzene was evaluated for chronic toxicity and carcinogenicity in Sprague-Dawley rats (60/sex/dose group) treated by gavage with dose levels of 0.0, 0.1, 0.7, and 5.0 mg/kg bw/day for at least 24 months. There was a dose-related increase in the levels of methemoglobinemia in the mid- and high-dose groups. High-dose rats displayed a slight increase in incidence of anemia (as indicated by slightly decreased hemoglobin, hematocrit, and erythrocyte values, and increased numbers of reticulocytes). High-dose males had elevated spleen weights and increased incidence and/or severity of accumulation of brown pigment. There was a dose-related increase in the incidence of interstitial cell tumors in the low- , mid-, and high-dose groups, as compared to the negative control group. The authors indicated that this effect was probably not biologically significant because the incidence of interstitial cell tumors in the negative control group was unusually low and the incidence of interstitial cell tumors in the treatment groups were within the range of historical negative controls. No increases in the incidences of other types of neoplasms were observed in treated rats. The treatment had no adverse effects with respect to mortality rate, clinical signs, body weights, food consumption, ophthalmoscopic examination, clinical chemistry, or urinalysis.
