Synthetic Receptor Affinities for the Entamoeba histolytica lectin

As the molecular biology of establishing a defined approach to lectin-carbohydrate interactions has evolved, the evolution of synthesis of complex carbohydrate structures has equally progressed. This study shows the possibilities of designing and synthesizing ligands capable of binding pathogenic adhesins with affinities high enough to afford capture of the pathogen within a fluid stream. A dissociation constant of 9.8 x 10(exp -10) M was achieved in BIAcore using lectin from Entamoeba histolyrica and synthesized GaINAc39BSA. The results compare favorably with in vitro membrane binding assays performed in Dr. Ron Schnaar's laboratory (Johns Hopkins School of Medicine). inhibition of binding, using galactose-like molecules, provides insight into the topography of the binding locale.
