In this work a comparative morphological and biochemical studies were conductedon T. spiralis isolated from pigs in Egypt and T. spiralis, T. nativa, T. Nelsoni and T. pseudospiralis reference species. Biochemical characterization of muscle larvae included isoenzyme analysis using 8 key enzymes: glucose 6-phosphate dehydrogenase (G6PD), malic enzyme (ME) acid phosphatase (ACP), phosphoglucomutase (PGM), glucose phosphate isomerase (GPI), mannose phosphate isomerase (MPI), fructokinase (FK) and hexokinase (HK) as well as molecular characterization using Southern lot analysis of Dra 1 digested genomic DNA. Results indicated characteristic protein bands for each species. G6PD was sufficient to discriminate among the four species as well as a unique 4.8 Kb rDNA band for T. spiralis (sensu stricto) in the pig isolates and T. spiralis reference strains. Biochemical and molecular characterization were more useful than morphological studies in the identification and differentiation of isolates and confirmed the identity of pig isolates to T. spiralis reference species.
