Determination of Carbamazepine with a RP-HPLC Method in Human Plasma Using Liquid- Liquid Extraction

摘要

Carbamazepine (CBZ) is an antiepileptic drug, which are prescribed as first-line drugs for the treatment of partial and generalized tonic-clonic epileptic seizures. A simple and sensitive Reversed Phase High Performance Liquid Chromatography (RP-HPLC) method for quantification of CBZ in human plasma was developed and validated. CBZ was extracted from human plasma with chloroform with greater recovery than 91.7% and separated using an isocratic mobile phase, on a reversed-phase C column. The effluent was monitored by UV detector at 220 nm and at a flow rate of 1.0 mL/min. The linearity range of proposed method was 0.5-25 μg/mL. The intraday and inter-day relative standard deviation and percent error values of the assay method were less than 9.14 % and the method was found to be precise, accurate, and specific during the study. The present method was selective enough to analyze CBZ in human plasma without any tedious sample clean-up procedure and was successfully applied for estimating the pharmacokinetic parameters of CBZ following oral administration of single dose of CBZ (Tegretol~R CR 200 (200 mg)) to six healthy volunteers.
