AbstractDominant transforming genes such asraf, myc, andras, alone or in combinations, have been shown to modify the radiation response of various cell types. Recent studies have shown that the status of the expression ofp53, the gene detected most frequently altered in human cancer, participates in the response of various cell types to DNA‐damaging agents, including ionizing radiation. We now report the radiation responses of three human cervical carcinoma cell lines expressing various endogenous levels of wild‐type or mutantp53. In order to modulate the levels of p53, cervical carcinoma cells were transfected with retroviral vectors in which a wild‐type full‐length humanp53cDNA was inserted in the sense or antisense orientation relative to the retroviral promoter sequences. Results showed that alterations in the endogenous levelsof p53did not result in consistent differences in the cellular responses to ionizing radiation. These data indicate that the level of wild‐type p53 is not a major participant in the mechanisms of response to ionizing radiation by these cervical carcinoma cells. © 1995 Wiley
