Tissue cultures isolated from different regions of pileus and spawn prepared on jowar grains gave higher yield as compared to cultures raised from other portion of fruit bodies on the same substrate. Bathu was the most inferior substrate for spawn preparation as it resulted in significantly lower yield irrespective of cultures. Cultures isolated from pileus middle (Pm) and stipe base (S,) exhibited minimum and maximum variation, respectively, regarding stipe length , stipe diameter and pileus diameter in the fruit bodies irrespective of different spawn substrates. The use of jowar grain as spawn substrate resulted in most desle morphological characteristics of fruit bodies irrespective of different cultures.Culture isolated from pileus centre (Pc) proved to be the highest yielder with fruit bodies having most desirable morphological features especially when jowar grain was used as spawn substrates.
