High cell metabolism above 0° C prevents prolonged storage of donor tissue for penetrating keratoplasty. Long-term preservation is only possible at low temperatures. In deep freeze storage, however, the addition of a protective agent such as dimethylsulphoxyddmsoor glycerol to the suspending medium is required so that cells are protected against damage during cooling to banking at and thawing from low temperatures
