Proteins in Cultured Human Fibroblasts and Keratinocytes

Human placenta extract has been used as a traditional drug to treat chronic hepatitis and menopausal disorders. Porcine placenta extract (PPE) is a nonprescription drug used as an analeptics and nutriment for treating weak constitution and physical exhaustion. The extract is also included in health foods and cosmetics, as it is expected to have an anti-aging effect. We therefore investigated the effects of PPE on the production of proteins associated with skin functions in cultured human dermal fibroblasts and keratinocytes. The expression levels of mRNAs for filaggrin (FLG), keratin10 (KIO), collagen, epidermal growth factor, fibroblast growth factors (FGFl and FGF2), and aquaporin-3 (AQP-3) in fibroblasts and keratinocytes treated with PPE were assessed by quantitative real-time PGR (qPCR). PPE stimulated the proliferation of cultured fibroblasts in a concentration-dependent maimer. qPCR analyses revealed that PPE potentiated the expression of FGFl and collagen type1(COLl) but not AQP-3. Production of procollagen type I C-peptide by fibroblasts was also increased by PPE. In contrast, PPE did not stimulate the proliferation of keratinocytes, even at higher concentrations, although it did increase the expression of mRNAs for FLG, KIO, and COLl in keratinocytes. The results obtained in this experiment indicate that PPE potentiates the expression of three proteins related to skin regeneration, suggesting it may be beneficial in cosmetics.
