To investigate the effects of silencing the human telomerase reverse transcriptase (hTERT) gene by RNAi on proliferation, apoptosis and expression of some related genes of colon cancer LoVo cell, the blank plasmid pYr1.1-Null and the interference plasmid pYr1.1-hTERT-shRNA were respectively transfected into LoVo cells. RT-qPCR was used to analyze the hTERT mRNA expression in the G418-selected transfected cells and normal LoVo cells. MTT and flow cytometry separately were applied to detect the proliferation and apoptosis of normal LoVo cells and transfected cells. RT-qPCR was applied to detect mRNA expression of EGFR, VEGFA, Caspase3, Bax and Bcl-2 gene after silencing hTERT gene. The hTERT mRNA expression level and cell proliferation rate in LoVo/shTERT were all lower than LoVo/Null and normal LoVo cell (P < 0.05). The cell apoptosis rate in LoVo/shTERT was higher than other cells (P < 0.05). The VEGFA mRNA expression level in LoVo/shTERT was lower than LoVo/Null and normal LoVo cell (P < 0.05) . The caspase3 mRNA expression level in LoVo/shTERT was higher than LoVo/Null and normal LoVo cell (P < 0.05). The EGFR, Bax and Bcl-2 mRNA expression levels showed no difference in each group (P > 0.05). Silencing of the hTERT gene by RNAi can block the hTERT mRNA expression, inhibit colon cancer LoVo cell proliferation and promote apoptosis.
