Quantitative analysis of L-lysine, L-threonine, L-homoserine and cobalamines in fermentation broth

L-Lysine, L-threonine, and L-homoserine have been separated from by-products (amino acids) in different biological fluids by TLC on Sorbfil plates with propan-2-ol-25% aqueous ammonia, 55 + 45 (v/v), propan-1-ol-acetone-water-25% aqueous ammonia, 25 + 25 + 7 + 6 (v/v), and propan-2-ol-ethyl acetate-25% aqueous ammonia-water, 20 + 20 + 1.5 + 25 (v/v) as mobile phases. A solution (1%) of ninhydrin in acetone was used as visualizing reagent and quantitation was performed after postchromatographic derivatization by dipping. Vitamin B_(12) and its derivatives were separated on aluminum oxide 60 F_(254) (type E) TLC plates with 2-methylpropan-l-ol-propan-2-ol-water, 30 + 20 + 20 (v/v) as mobile phase. Quantitation was achieved by conventional optical scanning and by videodensitometric scanning, and the results obtained were compared. Coefficients of variation < 5% were achieved by use videodensitometry, implying the suitability of this technique for rapid analysis of amino acids and vitamins.
