Thehighlyconservedplantaquaporins,knownasPlasmamembraneIntrinsicProteins(PIPs),arethemaingatewaysforcellmembranewaterexchange.YearsofresearchhavedescribedindetailthepropertiesofthePIP2subfamily.However,characterizingthePIP1subfamilyhasbeendifficultduetothefailuretolocalizetotheplasmamembrane.Inaddition,thediscoveryofthePIP1–PIP2interactionsuggestedthatPIP1aquaporinscouldberegulatedbyacomplexposttranslationalmechanismthatinvolvestrafficking,heteromerizationandfine‐tuningofchannelactivity.ThisreviewnotonlyconsiderstheevidenceandfindingsbutalsodiscussesthecomplexityofPIPaquaporins.ToestablishanewbenchmarkinPIPregulation,weproposetoconsiderPIP1–PIP2pairsasfunctionalunitsforthepurposeoffutureresearchintotheirphysiologicalroles...
