Flowcytometric and Immunohistochemical Observations on Cell Cycle and Surface Antigens inHuman Solid Tumours

Flowcytometric analysis was performed on 14 formalin preserved, histologically proven solid tumours, after treatingthem mechanically and enzymatically, obtaining bare nuclei. They were analyzed by Becton Dickinson Flowcytometerafter staining with propidium iodide. Non tumourous tissue was used as reference standard. The ABH(O) isoantigenwas studied for all the tumours by the specific red cell adherence (SRCA) test and intensity of reaction was gradedaccording to adhesion of erythocytes over the surface of the tissues. The quantitative DNA analysis showed that allthe tumours studied were aneuploid (Hypoploid) with 78% of cases showing two groups of population. Out of 14aneuploid tumours, 10 could be evaluated for S-phase fraction, which is highly predictive of disease free interval. Allwell differentiated tumours were SRCA positive and 25% of them were moderately positive and 75% were weaklypositive.While poorly differentiated tumours were all SRCA negative. Thus ABH(O) isoantigen was directly related toadvancing stage and degree of anaplasia. Hence, in addition to other clinical and pathological parameters, bothflowcytometric analysis of DNA and ABH(O) surface isoantigen are useful tests to predict recurrence and the overallsurvival of patients with solid tumours
