摘要

A new approach for developing plastome transformants was proposed and tested using Salpiglossis sinuata, family Solanaceae. Initially, chloroplasts of Salpiglossis sinuata were transformed in cybrid tobacco plants (Nicotiana tabacum L.) with S. sinuata plastome. Then, somatic hybridization was used to transfer S. sinuata chloroplasts from protoplasts of a N. tabacum (+S. sinuata) cybrid irradiated with super lethal doses into protoplasts of the wild-type of S. sinuata. These chloroplasts contain an insert of the aadA gene and have spectinomycin/streptomycin resistance. Finally plants that were morphologically similar to S. sinuata and resistant to spectinomycin/ streptomycin were developed. Molecular-biological analysis showed that the plants transformed plastome of S. sinuata. The analysis of telomeric and ribosomal fragments of nuclear DNA did not permit detection of tobacco-specific fragments. The analysis of mitochondrial DNA (mtDNA) showed the difference in mtDNA between the obtained plants and both parents.
