复方虫草提取物对实验性肺纤维化大鼠的影响

         

摘要

Objective To study the effects and interventional mechanisms of Cordyceps sinenisis extract on bleomycin-induced pulmonary fibrosis in rats. Methods The pulmonary fibrosis model of rat was established by intratracheal administration of bleomycin. The pulmonary index, pathomorphology, GSH, T-AOC, HYP, a-SMA, TGF-(3l protein and gene were assessed at the 7 d, 14 d, 28 d after drug administration. Lung samples collected were stained with hematoxylin-eosin (HE) staining and scored. BLM-prednisone group was positive control. Results Cordyceps sinenisis extract could alleviate BLM -induced pulmonary fibrosis in rats. The pulmonary index, the amount of HYP, the expression of a-SMA, TGF-(Jl and gene in lung tissue were significantly decreased in Cordyceps sinenisis extract group compared with those of control group (P <0. 01 or P <0.05) , While the activity of GSH and higher the level of T-AOC in lung tissue were enhanced in Cordyceps sinenisis extract group (P < 0. 01 or P < 0. 05 ). Conclusion Cordyceps sinenisis extract has an obvious relieving effect on rat pulmonary fibrosis, probably by the reinforcement of antioxidant systems, decreasing the expression of TGF-β1 and inhibiting the proliferation of lung myofibroblast.%目的 探讨复方虫草提取物对实验性肺纤维化大鼠的影响及其相关机制.方法 采取大鼠气管内注入博莱霉素的方法制备肺纤维化模型,分别在给予复方虫草提取物后第7、14、28天处死大鼠后测定肺系数、肺组织羟脯氨酸( HYP)含量、谷胱甘肽(GSH)的活力、总抗氧化能力(T-AOC)水平及α-平滑肌动蛋白(α-SMA)、转化生长因子β1(TGF-β1)蛋白和mRNA表达水平;取固定部位肺组织做病理组织学检查.以泼尼松作为阳性药对照观察.结果 复方虫草提取物能明显改善纤维化病变,降低肺系数、肺组织HYP含量及TGF-β1 mRNA、TGF-β1、α-SMA蛋白表达水平,尤其第28天显著(P<0.01或<0.05),且能明显升高GSH活力、T-AOC水平(P<O.01或0.05).结论复方虫草提取物可减轻肺泡炎症和肺纤维化的范围及程度,其作用机制可能与抗氧化作用和降低TGF-β1的表达进而抑制肺肌成纤维细胞的活化有关.
