本试验以甘蔗品种ROC22和粤糖93-159的愈伤组织和刚分化4周小苗为试验材料,初步确定了草铵膦在愈伤组织分化阶段和刚分化4周小苗阶段的筛选浓度。综合试验结果,适合草铵膦在ROC22和粤糖93-159愈伤组织分化阶段的筛选浓度为2.5µmol/L;适合草铵膦对ROC22刚分化4周小苗的筛选浓度为5.5µmol/L,适合草铵膦对粤糖93-159刚分化4周小苗的筛选浓度为6.6µmol/L。%In vitro glufosinate-ammonium selection regimes were respectively used to screen plants regeneration from callus and regenerated shoots (data recorded 4w after embryogenic callus were transferred to embryo germination medium) of ROC22 and YT93-159 for glufosinate-ammonium tolerance in this study. The high-severity treatments with high glufosinate-ammonium concentration which resulted in the necrosis of embryo germination or regenerated shoots were different for the different sugarcane cultivars. In conclusion, when the selection of herbicide-tolerant cell lines of sugarcane was applied to embryogenic callus, incorporating 2.5 µmol/L glufosinate-ammonium into culture medium of embryo germination from callus of the two cultivars was the optimum. When the selection of herbicide-tolerant cell lines of sugarcane was applied to shoot regeneration, incorporating 5.5 µmol/L glufosinate-ammonium into culture medium of regenerated shoots of ROC22 was the optimum, and incorporating 6.6 µmol/L glufosinate-ammonium into culture medium of regenerated shoots of YT93-159 was the optimum.
