十全大补汤总多糖的体外抗肿瘤作用

摘要

Objective To investigate the anti-tumor effects of total polysaccharide (TPS) extracted from Shiquan Dabu Decoction in vitro, so as to provide basis for its clinical application and research of anti-tumor drugs.Methods The human lung cancer cells A549, hepatoma cells Bel-7402, colorectal carcinoma cells HCT-8 and cervical carcinoma cells Hela cultured in vitro were treated with TPS of 200,100, 50 μg,/ml, the inhibitory rate of proliferation were measured by MTT assay at 24,48,72 h after the treatment.Results IR reached the highest in the A549 cells when treated with TPS of 200 μg/ml for 48 hs; IR in the other three kinds of cells increased gradually with the increasing of TPS concentration and prolonging of treating time, reached the highest at 200 μg/ml for 72 hs, which showed dose and time-dependent manner.Conclusions TPS could inhibit the growth of tumor in vitro; and hope to be used for the treatment of tumor as anti-tumor drugs.%目的 探讨十全大补汤总多糖(以下简称总多糖)的体外抗肿瘤作用,为其临床应用及抗肿瘤药物的研发提供依据.方法 以200、100、50 μg/ml浓度总多糖干预体外培养的人肺癌A549细胞株、人肝癌Bel-7402细胞株、人结肠癌HCT-8细胞株和人宫颈癌Hela细胞株.作用24、48、72 h时采用MTT法测定细胞增殖抑制率(IR).结果 200 μg/ml总多糖作用48 h时A549细胞的IR最高;总多糖对另3种细胞的作用均为随浓度及作用时间延长IR逐渐升高,以200 μg/ml、作用72 h时IR最高,呈剂量及时间依赖性.结论 总多糖可在体外抑制肿瘤细胞生长;其有望作为抗肿瘤药物用于临床.
