三阴型、Luminal型、HER2阳性型乳腺癌瘤组织中FA/BRCA通路基因表达对比观察

         

摘要

Objective To observe the differential expression of Fanconi anemia FA /BRCA pathway genes in different subtypes of breast cancer tissues and to explore the significance.Methods Ninety-three breast cancer tissues from inva-sive ductal carcinoma were collected during surgical resection.Among them, there were 20 cases of triple-negative breast cancer tissues,20 cases of Luminal breast cancer tissues,and 20 cases of HER2-positive breast cancer tissues.The num-ber of three types of breast cancer after neoadjuvant chemotherapy were 11,13 and 9 cases, respectively.The real-time PCR was used to detect the mRNA expression of genes(FANCA, FANCD2, FANCF, FANCI, BRCA1, and BRCA2) from the FA/BRCA pathway in the breast cancer tissues.Results The expression of BRCA1, BRCA2, FANCD2, and FANCI was significant higher in HER2-positive type breast cancer tissues than that of the other two subtype cancer tissues;the expression level of FANCA from Luminal breast cancer tissues was higher than those in the HER 2-positive type and tri-ple-negative type breast cancer tissues(all P<0.05).The expression of five genes from the FA/BRCA pathway except FANCF was low in neoadjuvant chemotherapeutic triple-negative breast cancer tissues.After neoadjuvant chemotherapy,the expression levels of BRCA1,BRCA2,FANCA,and FANCI from Luminal type breast cancer tissues were higher than those in the HER2-positive type and triple-negative type breast cancer tissues(all P<0.05).The expression levels of BRCA1, BRCA2,FANCD2,and FANCI in the triple-negative type and HER2-positive type patients who underwent neochemothera-py were lower than those in the patients who underwent the surgery directly,while the expression of FANCF remained high after neochemotherapy(all P<0.05).Conclusions Distinct expression pattern of FA-related genes in three types of breast cancer was found in our study.The role of FA/BRCA pathway gene in HER2-type breast cancer tissues is different from that in the other two subtypes.The high level expressions of BRCA 1,BRCA2,FANCA and FANCI may relate to the response of Luminal subtype to neoadjuvant chemotherapy.%目的 观察不同亚型乳腺癌组织中范科尼贫血(FA)/乳腺肿瘤易感基因(BRCA)通路基因的差异表达情况,并探讨其意义.方法 收集手术切除的女性患者乳腺癌组织标本共93例份,均为浸润性导管癌,其中直接手术的三阴型乳腺癌、Luminal型乳腺癌、HER2阳性型(HER2型)乳腺癌各20例,行新辅助化疗后的三型乳腺癌分别为11、13和9例.采用real-time PCR法检测乳腺癌组织中的FA/BRCA通路基因FANCA、FANCD2、FANCF、FANCI、BRCA1、BRCA2.结果 直接手术的HER2型乳腺癌组织中BRCA1、BRCA2、FANCD2、FANCI基因表达量均高于三阴型和Luminal型,Luminal型乳腺癌组织中FANCA基因表达量高于HER2型和三阴型(P均<0.05).新辅助化疗后的三阴型乳腺癌组织中除FANCF外其余5个基因表达量均非常低,新辅助化疗后的Luminal型乳腺癌组织中BRCA1、BRCA2、FANCA、FANCI基因表达均高于三阴型和HER2型(P均<0.05).新辅助化疗后的三阴型、HER2型乳腺癌组织中BRCA1、BRCA2、FANCD2、FANCI基因表达量低于同型直接手术者(P均<0.05),新辅助化疗后三个亚型乳腺癌组织中FANCF的表达水平高于直接手术者(P均<0.05).结论 三种亚型乳腺癌组织中FA/BRCA通路基因表达差异较大;FA/BRCA通路基因在HER2型乳腺癌与其他两亚型乳腺癌中作用差异较大;BRCA1、BRCA2、FANCA、FANCI基因高表达可能与Luminal型乳腺癌患者新辅助化疗预后有关.
