RNA干扰技术的研究进展

         

摘要

RNA干扰(RNAi)指由dsRNA或siRNA诱发的转录后基因沉默现象,其机制是通过与靶序列特定位点互补配对,通过阻碍靶基因的翻译或者诱导mRNA降解来抑制基因表达,是一项高效率、强特异性的基因沉默技术。 RNAi技术不仅在基础研究方面成为基因功能和蛋白功能研究的工具,也广泛的应用于临床研究,在肿瘤、病毒性疾病以及其他疾病的治疗方面显示了广阔的前景。%RNA interference (RNAi) refers to the post-transcriptional gene silencing induced by dsRNA or siRNA. The mechanism is that the dsRNA or siRNA was pairing complementarily to the specific sites of the target sequence, which would hinder the transcription of the target se-quence or induce mRNA degradation to inhibit gene expression. RNAi was a high-efficiency, high-specific gene silencing technology, which is not only a important tool in the fundamental re-search, but also widely used in clinical studies in the treatment of cancer, viral diseases and other diseases.
