目的:筛选TTC法测定独活种子生活力的最佳条件,并明确生活力与发芽率的相关性.方法:比较不同浓度、温度、时间条件下染色的效果;以15份不同来源独活种子为材料,进行生活力测定和发芽率检测.结果:TTC法测定独活种子生活力的最适条件是45℃恒温条件下在1%的TTC溶液中染色50 min;独活种子生活力(X)与发芽率(Y)回归方程为Y =0.7326X-0.09%.结论:45℃恒温条件下在1%的TTC溶液中染色50 min,是测定独活种子生活力的适宜条件,其测得的生活力与发芽率呈显著的正相关.%Objective:To screen the optimized methods for detection seed viability of Angelica pubescens seed,and determine the relationship between seed viability and germination rate.Method:The effect of dyeing in different concentrations,temperature and time conditions was compared.Use 15 seeds from different sources as a material,the determination of viability and germination rate were tested.Results:The optimum condition of determination seed viability using TIC is soaking seeds in the 1% TTC solution for 50 minutes under the condition of 45 ℃.The regression equation was found out between seed viability (X) and germination rate (Y),which was Y =0.7326 X-0.09 % . Conclusion:The optimum condition of determination seed viability using TIC is soaking seeds in the 1% TTC solution for 50 minutes under the condition of 45 ℃.Furthermore,seed viability was significant related with germination rate of Radix Angelica pubescens seed.
