去整合素-基质金属蛋白酶在肿瘤细胞中的作用研究进展

     

摘要

去整合素-金属蛋白酶(ADAM)家族是许多细胞外基质(ECM)的成分。根据其结构的不同分为膜锚定型和分泌型,这些分子在各种生物活动(如细胞黏附、细胞融合、细胞迁移、膜蛋白脱落和蛋白水解)中发挥作用。一些ADAMs成员在恶性肿瘤的病理过程中发挥作用。 ADAMs的活性受基因表达、胞质和周围细胞的调控,酶原和抑制因子也参与调控。尽管现在还不清楚具体的作用机制,但是它们中大多数具有调控生长因子活性,整合、诱导细胞增殖和侵袭的作用。%Adisintegrin and metalloproteinases (ADAM) family,which is divided into membrane anchor type and secretory type depending on their structure,are main extracellular matrix(ECM) components.These molecules play a role in a variety of biological activities ,such as cell adhesion,fusion,migration,membrane-protein shedding and protein hydrolysis.Some ADAM members play important roles in the pathogenesis of malignancies.ADAMs activity is regulated by gene expression,cytoplasm and surrounding cells as well as proenzymes and inhibitors.Although the specific mechanism of actions is unknown,most of them have growth factor regulatory activity and effects of integration and induction of cell proliferation and invasion .
