自噬是一种高度保守的溶酶体降解的过程,其能降解细胞中受损的细胞器和错误折叠蛋白,以维持细胞内环境稳定,自噬对肿瘤具有双重作用,其既能为肿瘤细胞提供营养和保护,又可抑制或杀伤肿瘤细胞,而Beclin1是重要的自噬相关基因,其在肿瘤的发生发展中可调节自噬状态,其表达改变可影响肿瘤发生发展,并与化疗耐药相关.了解Beclin1的结构、作用机制、影响因素以及与消化道肿瘤的关系,将会为消化道肿瘤的检测、诊断、治疗、预后提供新策略.%Autophagy,a highly conserved lysosomal degradation process,can degrade misfolded proteins and impaired organelles to stabilize intracellular environment in cells.Autophagy has a dual role in cancer,which can not only provide nutrition and protection for tumor cells,but also inhibit the growth of tumor cells or kill them.A key autophagy-related gene Beclin1 may regulate autophagic state in cancer formation and progression.Changes in its expression can affect occurrence and development of tumor,and it is related to chemotherapeutic drug resistance.Better understanding of Beclin1′s structure,function,influencing factors and relations with gastrointestinal tumor,may provide new strategies for the detection,diagnosis,treatment and prognosis of gastrointestinal tumor.
