华南忍冬不同器官中黄酮类物质的定位与定量及苯丙氨酸解氨酶活性分析

         

摘要

Laser scanning confocal microscopy ( LSCM) was used to investigate the localization and rela-tive quantity of flavonoids in stem, leaf and flower of Lonicera confusa DC.Simultaneously, the phenyla-lanine ammonialyase ( PAL) activity was determined and the correlation between flavonoid contents and PAL activities was further analyzed in this paper.The results showed that the flavonoids localized ubiqui-tously in epidermal cells, parenchyma cells and vascular bundle of the stems, leaves and flowers of Loni-cera confusa.The flavonoids accumulated mainly in the xylem vessel walls of stems and leaves, and the epidermal hair of flowers.The statistical analysis showed that the flavonoid contents varied among diffe-rent organs and different parts of the same organ in Lonicera confusa, with much higher flavonoid contents in corolla-anther and calyx tube-ovary than those in stem and leaf.Within the corolla tube, the lowest content of flavonoids localized in the middle part of corolla tube.The PAL activities varied in accord with the flavonoid contents in stems, leaves and flowers of Lonicera confusa, indicating that the synthesis of flavonoids is closely related with the PAL activity in Lonicera confusa.%采用激光共聚焦显微技术对华南忍冬( Lonicera confusa DC.)的茎、叶和花蕾中的黄酮类物质进行了组织定位和相对定量,同时对其苯丙氨酸解氨酶( Phenylalanine ammonialyase, PAL )的活性进行了测定,并对黄酮类物质含量与PAL活性的相关性进行了分析.结果显示:在华南忍冬的茎、叶和花蕾的表皮细胞、维管束和薄壁细胞都有黄酮类物质分布,其中以茎和叶的木质部导管壁以及花蕾表皮毛黄酮类物质累积较多;在华南忍冬不同器官和同一器官的不同部位之间黄酮类物质的含量差异显著,其中花蕾的花冠-花药和花萼筒-子房的黄酮类物质含量均高于茎和叶,而花蕾中花冠筒中部的黄酮类物质含量最低;华南忍冬茎、叶和花蕾的PAL活性与黄酮含量变化趋势一致,意味着华南忍冬黄酮类物质的合成与PAL活性密切相关.
