Objective To study the effects of breviscapine on the expression of interleukin-1β(IL-1β) in lung tissues with left heart ischemic reperfusion of rabbits. Methods Sixty New Zealand rabbits were randomly divided into the model group and the therapy group. The rabbits in model group copied the model of left heart ischemia-reperfusion that was made by ligaturing and loosing the anterior descending branch. The therapy group not only copied the model but also cured by breviscapine injection with 10 mg · Kg-1. Then the rabbits were sacrificed,phrenic nerve discharge curve,the levels of IL-1β in peripheral blood and bronchoalveolar lavage fluid, the expression of IL-1β(5 in lung tissue,the changes of ultrastructural organization were observed after ischemia for 30 minutes,reperfusion for 20 minutes and 40 minutes. Results The duration and amplitude of phrenic nerves discharge curves in therapy group were higher than model group( P <0.05) ,the levels of IL-1β in peripheral blood and bronchoalveolar lavage fluid and lung tissue in therapy group were all lower than those in model group(p<0.05). By electron microscopy, type I and type Ⅱ alveolar epithelial cell in model group showed the acute inflammatory lesions invaria-nce change,but the damage in therapy group was little. Conclusion Breviscapine may restraint the rabitts' acute lung injury following left heart ischemia-reperfusion through reducing expression of IL-1β in lung.%目的 探讨灯盏花素对左心缺血再灌注后肺组织白细胞介素-1β(IL-1β)表达的影响.方法 60只成年健康新西兰家兔随机分为模型组和治疗组.2组家兔均结扎及再通左冠状动脉前降支复制左心缺血-再灌注模型,治疗组在缺血后10 min经静脉给予灯盏花素注射液10 mg·kg-1.观察缺血30 min、再灌注20、40min时2组家兔膈神经放电曲线、外周血和支气管肺泡灌洗液(BALF)中IL-1β水平、肺组织细胞IL-1β表达情况.结果 治疗组各时间点膈神经放电幅度和持续时间明显比模型组增大、延长(P<0.05);治疗组外周血、BALF和肺组织细胞中IL-1β水平均低于模型组(P<0.05).结论 灯盏花素可减少IL-1β的表达,减轻因左心缺血再灌注而导致的急性肺损伤.
