石榴皮多酚提取物亚急性毒性实验研究

         

摘要

Objective To investigate the subacute toxicity of Pomegranate Peel Polyphenols extract. Methods The mice were divided into blank control group,1600,800,and 500mg/kg3 dose group.Continuous feeding for 30 days,2/3 animals were dissected after the last administration for 24 h.The rest mice were sacrificed after 2 weeks′recovery of the observation.Blood routine and biochemical indexes were determined by the eyeball blood and the heart,liver and spleen,lung and kidney of mice were taken out and weighed to observe the pathological changes of the organs.At the end of the drug administration and and the end of the recovery period,the examination was taken once respectively.Results At the end of the dosing period,there was no significant difference in body weight and food utilization rate of mice in each group compared with the control group (P >0.05).Compared with the control group,the difference was statistically significant in RBC value of high and middle dose groups (P <0.05)and WBC value in high, medium and low dose group (P <0.01)and Neu,Lym in medium and low dose group of mice;The Eos value of the female mice in the high dose group and the middle dose group was significantly different from that in the control group (P <0.05),and the Eos value of the middle and low dose male mice was significantly different from that of the control group (P <0.05).TG and UREA values of mice in the low,mid-dle and low dose groups were significantly different from that of the control group (P <0.05).The liver coefficient of female mice in blank group was 5.2,and the liver coefficient is high,which in high,medium and low dose group were 4.8,4.3,4.4,respectively.While the liver coefficient of male mice in blank group was 5.2,the liver coefficient is higher,which in the high,medium low dose group were 5.1,4.6,4.5,respectively;There was a significant difference between the high dose group and the control group in kidney coefficient (P < 0.05).At the end of the recovery period,there was no significant difference in body weight,food utilization rate and blood indexes of mice in each group compared with the control group (P>0.05).Compared with the control group,TG value in high dose group was statistically significant differ-ence (P <0.05).In the recovery period,organ coefficient of mice in high dose group and the blank was statistically significant (P <0.05)compared to the control group.The liver index of female mice in blank group was 4.8,which in high and middle dose groups were 4.7,5.1,respectively.The liver index of male mice in blank the group of 4.3,which in high and middle dose groups were 5.5,5.0,respectively.Conclusion Pomegranate peel extract in the test results showed no significant toxicity,but high dose or unrea-sonable use will also have adverse effects on the health of mice.%目的 研究石榴皮多酚提取物的亚急性毒性作用.方法 将120只小鼠随机分为空白对照组和石榴皮多酚提取物高、中、低剂量组,给药剂量依次为1600、800、500 mg/kg体质量,每组30只.连续喂养30 d,末次给药后24 h剖杀2/3小鼠,剩余小鼠恢复观察2 w后剖杀,摘眼球采血作血常规及生化指标测定,取出心、肝、脾、肺、肾称重,观察小鼠各器官、病理变化情况.给药结束和恢复期结束时各检查1次,共计2次.对恢复期小鼠的肝脏进行病理组织学检查.结果 给药期结束时,各组小鼠生长活动正常,各剂量组小鼠体质量变化、食物利用率与空白对照组比较,差异均无统计学意义(P>0.05);小鼠血液指标高、中剂量组小鼠RBC值与空白对照组比较,差异有统计学意义(P<0.05);高、中、低剂量组小鼠WBC值与空白对照组比较,差异有统计学意义(P<0.01);中、低剂量组小鼠Neu、Lym值与空白对照组比较,差异有统计学意义(P<0.05);高、中剂量组雌鼠的Eos值与空白对照组比较,差异有统计学意义(P<0.05),中、低剂量组雄鼠的Eos值与空白对照组比较,差异有统计学意义(P<0.05).高、中、低剂量组小鼠血液生化指标TG、UREA值与空白对照组比较,差异有统计学意义(P<0.05).各剂量组小鼠肝脏系数与空白对照组比较,差异有统计学意义(P<0.01);雌鼠空白组的肝脏系数为5.2,高、中、低剂量组小鼠的肝脏系数依次为4.8、4.3、4.4,雄鼠空白组的肝脏系数为5.2,高、中、低剂量组小鼠的肝脏系数依次为5.1、4.6、4.5;高剂量组小鼠的肾脏系数与空白对照组比较差异有统计学意义(P<0.05).恢复期结束时,各组小鼠生长活动正常,各剂量组小鼠体质量变化、食物利用率与空白对照组比较,差异均无统计学意义(P>0.05);小鼠血液指标差异无统计学意义,高剂量组小鼠血液生化指标TG值与空白对照组比较,差异有统计学意义(P<0.05),恢复期高、中剂量组小鼠的脏器系数与空白对照组比较,差异有统计学意义(P<0.05),雌鼠空白组的肝脏系数为4.8,高、中剂量组小鼠的脏器系数分别为4.7、5.1,雄鼠空白组的肝脏系数为4.3,高、中剂量组小鼠的脏器系数分别为5.5、5.0.结论 石榴皮多酚提取物未表现出明显毒性,高剂量或者不合理用药均会对小鼠健康产生不利影响.
