目的 探讨两色金鸡菊乙酸乙酯提取物对高糖诱导大鼠肾小球系膜糖尿病肾病(DN)细胞模型的影响及其作用机理.方法 通过高糖诱导大鼠肾小球系膜细胞建立体外DN细胞模型,应用两色金鸡菊乙酸乙酯提取物不同浓度对细胞模型进行干预后,分为正常组、高糖组及高糖+两色金鸡菊乙酸乙酯提取物不同剂量(25、50、100、150μg/mL)组共6组.采用Real time-PCR法、Western blot法,从转录水平及蛋白表达水平检测其对细胞中炎性因子单核细胞趋化蛋白-1(MCP-1)、细胞间黏附分子-1(ICAM-1)以及对NF-κB信号通路中p-65表达的影响.结果 两色金鸡菊乙酸乙酯提取物能明显抑制高糖诱导大鼠肾小球系膜细胞中炎症因子MCP-1 mRNA和蛋白表达水平,以及ICAM-1的mRNA表达水平.两色金鸡菊乙酸乙酯提取物能明显抑制高糖诱导大鼠肾小球细胞中p-65蛋白的表达.结论 两色金鸡菊乙酸乙酯提取物可能通过NF-κB信号通路抑制DN细胞模型的炎症反应.%Objective The aim of our study was to discuss the pharmacological effects of ethyl acetate extract of Coreopsis tinctoria Nutt.on high glucose-induced rat glomerular mesangial cell.Methods Rat glomerular mesangial cell were divided into six groups which were normal cell group,high glucose induced group,groups of ethyl acetate extract of Coreopsis tinctoria Nutt.in different concentrations(25,50,100, 150μg/mL).Real time-PCR method and Western method were used to detect the effects of ethyl acetate extract of Coreopsis tinctoria Nutt on the expression of inflammatory factor MCP-1 ,ICAM-1 and the expression of NF-κB in the signal pathway of transcription factor and protein level.NF-κB signaling pathway was detected to reveal the mechanism of anti-inflammatory effect of Ethyl acetate extract of Coreopsis tinctoria Nutt.Results In vitro,ethyl acetate extract of Coreopsis tinctoria Nutt.significantly decrease mRNA and protein expression of MCP-1 ,ICAM-1 and p-65 induced by high glucose in rat glomerular mesangial cell.Conclusion Ethyl acetate extract of Coreopsis tinctoria Nutt.inhibits inflammatory effect of high glucose-induced rat glomerular mesangial cell.The possible anti-inflammation mechanism is by NF-κB signaling pathway.
