灸预处理对急性胃黏膜损伤大鼠胃黏膜微循环屏障相关因子作用的影响

摘要

目的 观察灸预处理对急性胃黏膜损伤大鼠胃组织一氧化氮(nitric oxide,NO)和内皮素(endothelin,ET)的影响,探讨灸预处理对大鼠胃黏膜保护中胃黏膜微循环屏障相关因子的调节作用.方法 将 40 只 SPF级健康SD大鼠随机分为 4 组,正常组、模型组、灸预处理组和灸预处理非穴位组.首先灸预处理组选取足三里、中脘进行艾条温和灸 7 d,灸预处理非穴位组选取非穴位的对照点行相同灸治.之后采用无水乙醇结合阿司匹林混悬液灌胃法进行造模.以 Guth法计算胃黏膜损伤指数(UI),酶免法检测胃黏膜组织匀浆中 NO、ET 的含量.结果 与正常组比较,模型组大鼠 UI指数显著增加,胃组织 NO含量明显降低,ET含量明显降低(P ＜0.01);与灸预处理非穴位组比较,灸预处理组大鼠 UI指数显著降低(P ＜0.05),胃组织NO含量明显降低(P ＜0.05),ET含量明显降低(P ＜0.01).结论 灸预处理足三里、中脘可减轻无水乙醇结合阿司匹林混悬液灌胃造成的大鼠急性胃黏膜损伤,可能是通过上调胃黏膜微循环屏障相关保护因子NO,下调其损伤因子ET,以达到加强胃黏膜微循环屏障,抵抗胃黏膜损伤的作用.%Objective To investigate the protective effect of moxibustion pretreatment on gastric mucosa of rats and the regulatory effect on gastric mucosa microcirculation barrier related factors,by observe the effects of acupuncture and moxibustion preconditioning on contents of nitric oxide(NO),and endothelin(ET)in rats with gastric mucosal injury.Methods 40 SPF SD rats were randomly divided into normal,model,acupuncture pretreatment(AP),moxibustion preconditioning(MP)groups.After 7 days of acu-puncture and moxibustion preconditioning with Zusanli(ST 36),Zhongwan(CV 12),the rats were given anhydrous ethanol combined with aspirin suspension by gavage to make the gastric mucosal injury model.The gastric mucosal injury index(UI)was evaluated by Guth method,and the determination of NO and ET in gastric mucosa homogenate by enzyme immunoassay.Results Compared with the AP group,the UI index increased significantly(P＜0.05),while the contents of NO decreased markedly(P＜0.05)and ET in-creased markedly(P＜0.01)in the MP group.Conclusion Acupuncture and moxibustion preconditioning can strengthen the microcirculation of gastric mucosaalleviating,and resistance the gastric mucosal damage,by up-reg-ulating the protective factor NO of gastric mucosal microcirculation,and down-regulating its damage factor ET.The effect of moxibustion preconditioning obviously superior to the acupuncture pretreatment.
