HepG2 细胞谷胱甘肽含量和谷胱甘肽转移酶活力的诱导

         

摘要

Objective : To develop a kind of HepG2 variant with high GSH content and GST activity. Methods: HepG2 cells were pulse treated with 0.1, 0.3, 0.5 mg/L CDDP for 1h every week, respectively, Twelve months later, three CDDP-resistant cell lines (HepG2/CDDP0.1, HepG2/CDDP0.3, HepG2/CDDP0.5 ) were obtained. GSH content and GST activity were measured. The results were statistically analyzed by the method of t - test. Results: There were two - fold elevated of GSH and GST exist In HepG2/CDDP0.1, HepG2/CDDP0.3, HepG2/CDDP0.5 comparing to the parental HepG2. And there were significant discrepancies of GSH contains between HepG2 and HepG2/CDDP 0.3, HepG2/CDDP 0.5. Conclusion: All evidence suggest that GSH and GST can be induceed under the treatment of CDDP.%目的:建立一种具有较高 GSH 含量和 GST 活力的肝细胞系。方法:将 HepG2 细胞在 0.1,0.3,0.5 mg/L 浓度的 CDDP 间歇作用下传代 12 个月后得到三种稳定的 CDDP 耐药细胞系 HepG2/CDDP0.1,HepG2/CDDP0.3,HepG2/CDDP0.5 细胞的 GSH 含量和 GST 活力,结果用显著性 t 检验方法作统计学处理。结果:诱导刺激后的子代 HepG2/CDDP0.1,HepG2/CDDP0.3,HepG2/CDDP0.5细胞的 GSH 浓度和 GST 活力较亲代有成倍的增长,且 HepG2/CDDP0.3,HepG2/CDDP0.5 细胞的GSH 浓度确较亲代有显著性差异。结论:细胞 GSH 含量、GST 活力是可以诱导的,此种以 CDDP 刺激HepG2 产生高 GSH 和 GST 的方法是可行的。
