阿特拉津对雄性大鼠骨髓微核率及肾脏和脾脏组织学变化的影响

         

摘要

试验旨在探讨阿特拉津对雄性大鼠骨髓微核率及肾脏和脾脏组织学变化的影响.选取60只4周龄雄性大鼠,随机分成4组(对照组和低、中、高3个剂量组),每组15只.剂量组大鼠分别按每千克体重1/32LD50(55.625mg)、1/16LD50(111.5mg)、1/8LD50(225.5mg)于试验的0,30,60d腹腔注射阿特拉津,并于试验的30,60,90d处死大鼠,分别采集骨髓、肾脏和脾脏等组织,检测骨髓微核率、肾脏和脾脏的组织学变化等指标.结果表明:长时间应用阿特拉津,骨髓微核率升高,低、中、高剂量组分别比对照组高出17.14%、133.33%和119.52%;而高剂量组以60d时最高,分别比30d和90d高出118.65%和55.10%;肾脏组织则出现增生性和退行性病变、脾脏组织脂肪变性,从而说明,长时间应用阿特拉津可能导致雄性大鼠出现一定的遗传毒性反应,并导致肾脏和脾脏组织病变.%The purpose of this study was to investigate the effect of atrazine on the micronucleus rates of bone marrow and the histological changes of kidney and spleen in male rats.Four-week-old 60 male rats were randomly divided into 4 groups (control,low,middle,and high dose groups),15 in each group.In dose groups,rats were afforded atrazine by intraperitoneal injection according to 1/32 LD50 (55.625mg),1/16 LDS0 (111.Smg) and 1/8 LD50 (225.5mg) per kg weight on 0 day,30d,60d,respectively.The rats were killed at the 30d,60d and 90d,then bone marrow,kidney and spleen tissue samples were collected to detect the bone marrow micronucleus rates and histological changes of kidney and spleen.The results showed that the application of atrazine for longer time,micromucleus rates of bone marrow increased.Low,medium,high dose groups'micromucleus rates were respectively 17.14%,133.33%,119.52% higher than that of the control group.The high dose group was the highest in 60d,which was respectively higher 118.65%,55.10% than that of 30d and 90d.The kidney tissue presented proliferative and degenerative changes,spleen tissue presented fatty degeneration,which showed that the application prolonged of atrazine may lead to male rats appeared certain genetic toxicity and kidney and spleen tissue lesions.
