黑果腺肋花楸总黄酮提取工艺优化及抗氧化活性研究

         

摘要

Aronia melanocarpa Elliot fruit is purple and black berries.Its fruit contains rich flavonoids and other substances,and it has high value of the research.In this paper,the extraction technology of flavonoids from A.melanocarpa Elliot was optimized by enzymatic method,and the antioxidant ability was studied by using single factor orthogonal experiment.The results showed that the optimum parameters as following:the pectinase concentration 10 mg/mL,the ethanol concentration 15%,1 ∶ 23 (g/mL) at 60 ℃ for 90 min;in this condition Aronia flavonoids yield was 373 mg/g.Antioxidant experiments showed that:flavonoid concentration was ositively related to antioxidant capacity,DPPH · clearance rate's,the superoxide anion clearance rate's and the hydroxyl radical clearance rate's EC50 values were 5.05 mg/mL,2.31 mg/mL and 3.40 mg/mL,respectively.Superoxide anion clearance rate> hydroxyl radical clearance rate > DPPH · clearance rate and flavonoids from A.melanocarpa Elliot had strong antioxidant capacity.%黑果腺肋花楸(Aronia melanocarpa Elliot)果实为浆果,果实内含有丰富的黄酮类等物质,具有很高的研究价值.本文以黑果腺肋花揪为原料,采用单因素结合正交的实验方法,优化了酶法辅助提取黑果腺肋花揪黄酮类化合物的提取工艺,并对其抗氧化能力进行了研究.结果表明,黑果腺肋花揪总黄酮类的最佳工艺参数为:果胶酶浓度10 mg/mL,乙醇浓度15%,料液比1∶23 (g/mL),提取温度60℃,提取时间1.5h.在此条件下,黄酮的得率为373 mg/g.抗氧化实验结果表明:黄酮浓度与抗氧化能力呈正相关关系,DPPH自由基清除率EC50=5.05 mg/mL,超氧阴离子清除率EC50=2.31 mg/mL,羟自由基清除率EC50=3.40 mg/mL,超氧阴离子清除能力>羟自由基>DPPH自由基,说明黑果腺肋花楸中总黄酮类有良好的抗氧化活性.
