Objective To analyze the serum protein/peptides spectrum characteristics of healthy adult and to provide basic information for human serum proteomic study.Methods Serum protein/peptides of 30 healthy adult linked to weak cation exchange protein chip (WCX2 bead) were detected by surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS).And the experimental data,inclucling the kinds,range of relative molecular mass,abundance measured in relative strength and individual variety,were analyzed descriptively.Results 947 kinds of protein/peptides,whose relative molecular masses range from 1 059 to 10 265were found out and there were 34 ～ 108 kinds of protein/peptides in various individuals.The abundance of these serum protein/peptides were in the range of 95 to 35 419.As to individual variety,570,154,71,36,32,26,18,16,14,5,2 and 3 kinds of serum protein/peptides existed in 1,2,3,4,5,6,7,8,9,10,11,14 subjects,respectively.Conclusions WCX2 bead can only capture these human serum protein/peptides of which the quantity,the abundance and the relative molecular mass is less,smaller and lower than that of being.Furthermore,they present significant individual variety among subjects.%目的:分析健康成年人血清蛋白质/多肽谱特点,为人类血清蛋白质组研究提供基础性资料.方法:应用弱阳离子交换蛋白质芯片(WCX2)磁珠处理30例健康成年人血清,通过表面增强激光解吸-电离飞行时间质谱(SELDI-TOF-MS)技术检测血清蛋白质/多肽谱,描述性分析其种类、分子质量范围、丰度(用相对强度计量)、个体性差异等.结果:共检测到947种蛋白质/多肽,不同个体其种类数量在34～108不等,相对分子质量范围为1 059～ 10265,表达丰度在95 ～ 35 419之间;分别有570、154、71、36、32、26、18、16、14、5、2和3种相应血清蛋白质/多肽在1、2、3、4、5、6、7、8、9、10、11、14例个体血清中存在.结论:WCX2磁珠仅能捕获到数量较少的低丰度、低分子量人血清蛋白/多肽,在受检人群中这些蛋白/多肽呈现高度的个体差异性.
