枸杞多糖减轻长波紫外线辐射致人皮肤成纤维细胞损伤的超微结构观察

         

摘要

Objective To observe the ultrastructural effects of Lycium Barbarum Polysaccharides on fibroblast in vitro irradiated by ultraviolet A (UVA). Methods Taking the primary cultured fibroblast as objects, the fibroblast was irradiated by UVA (irradiation intensity : 2.4J/cm2). The fibroblast was randomly divided into 5 groups,control group,UVA radiated group and three LBP protective groups, 0. 2mg · mL-1 LBP,0.4 mg · mL~' LBP and 0. 8mg · mL-1 LBP group respectively, electronic microscope was used to observe the ultra-structure of the cells. Results After the fibroblasts were irradiated by UVA (irradiation intensity: 2. 4J/ cm2 ) , the ultrastructure changed obviously, the nuclear membrane unintegrated, and it showed the gathering of heterochromatins in the nucleus, mitochondrial swelling, vacuolization, crista fragmentation and disappearing, dilatation of rough endoplasmic reticulum. Preconditioning of LBP decreased such damage UVA radiation on mitochondria! And endoplasmic reticulum. Conclusion UVA radiation can change ultrastructure of fibroblast, LBP can decrease such damage of fibroblast irradiated by UVA%目的 观察枸杞多糖(lycium barbarum polysaccharides,LBP)对长波紫外线(UVA)辐射损伤体外培养人皮肤成纤维细胞超微结构的影响.方法 以体外培养的原代人皮肤成纤维细胞为实验材料,采用总剂量为2.4 J/cm2的UVA照射细胞,实验分为对照组、紫外线照射模型组和三个不同浓度LBP保护组(0.2mg· mL-1LBP+UVA,0.4mg·mL-LBP+UVA,UVA +0.8mg· mL-1 LBP+UVA),应用透射电镜观察各组细胞超微结构的变化.结果 UVA辐照后细胞超微结构发生明显的变化,主要表现为核膜不连续,核内出现团块状染色质,胞浆内线粒体肿胀,空化明显,嵴断裂、溶解,粗面内质网扩张.枸杞多糖预处理可以减轻对线粒体,内质网等细胞器的损伤;且随LBP剂量的升高,辐射损伤程度逐渐减轻.结论 UVA照射可使体外培养的人皮肤成纤维细胞超微结构改变,LBP可以减轻UVA所致人皮肤成纤维细胞的损伤.
