2-BFI对EAE大鼠的脊髓TGF-β和IL-10免疫组化表达的影响

         

摘要

目的 探讨2-BFI对实验性自身免疫性脑脊髓炎大鼠脊髓转化生长因子-β(TGF-β)和白介素-10(IL-10)表达的影响.方法 50只雌性SD大鼠根据随机数字表法随机分为5组:对照组、EAE组、低剂量2-BFI干预组(1.5mg/kg)、中剂量2-BFI干预组(3mg/kg)和高剂量2-BFI干预组(6mg/kg),每组10只,采用豚鼠脊髓匀浆(GPSCH)免疫诱导SD大鼠建立EAE模型,观察每组大鼠发病情况并进行临床症状评分,采用常规HE染色和LFB髓鞘染色观察中枢神经系统(CNS)的病理变化,免疫组化法检测TGF-β和IL-10表达.结果 与EAE组比较,大、中、小剂量2-BFI干预组大鼠EAE发生率下降,临床症状减轻,潜伏期延长,CNS内炎性细胞浸润减少,但仅中等剂量2-BFI在EAE临床症状和CNS内炎性细胞浸润方面差异有统计学意义(P<0.05);与EAE组比较,中等剂量2-BFI干预组脊髓中TGF-β和IL-10表达明显增多,差异有统计学意义(P<0.05).结论 中等剂量2-BFI对EAE大鼠的神经保护作用最佳,其作用推测与上调TGF-β和IL-10的表达有关.%Objective To explore the effect of 2-BFI on transforming growth factor-β (TGF-β) and interleukin-10 (IL-10)in spinal cord of experimental autoimmune encephalomyelitis (EAE) in rats.Methods Fifty female Sprague Dawley rats were randomly divided into five groups according to randomly digital table:control group(n =10),EAE model group(n =10),2-BFI 1.5 mg/kg treatment group (n =10),2-BFI 3 mg/kg treatment group (n =10) and 2-BFI 6mg/kg treatment group (n =10).The model of EAE in SD rats was induced by Guinea pigs spinal cord homogenate(GPSCH).The severity of EAE was scored according to the signs and symptoms.CNS tissues were stained with Hematoxylin-Eosin.Then the pathological changes of CNS were observed.Pathological changes were observed with the aid of Hematoxylin-eosin staining and Fast blue luxol dyeing.The levels of TGF-β and IL-10 were detected by immunohistochemistry.Results Compared with EAE group,the rats in the low,medium and high dose 2-BFI treatment group had lower incidence of disease,prolonged latency,decreased CNS inflammation and demyelination.But only medium dose 2-BFI treatment group had significantly lower alleviated clinical symptom and decreased CNS inflammatory cell infiltration(P < 0.05),higher level of TGF-β and IL -10(P < 0.05).Conclusion 2-BFI at medium dose of 3mg/kg has a beneficial neuroprotective effect on EAE.Its role might be supposedly related to the increased expression of TGF-β and IL-10.
