Objective To explore the enriching method for the total flavonoids from male flowers of EucommiaulmoidesOliv with polyamide-macroporous resin .Methods Polyamide adsorbed with sample was washed with water first to eliminate the impurities and then was loaded on the top of macroporous resin .The total flavonoids were enriched by eluting with ethanol .Results D101 resin combined with polyamide could effectively enrich the total flavonoids .The preferred condition for the procedure was as following :the weight of polyamide and dry D 101 resin was three and ten times of the extract weight( 1 ∶ 3 and 1 ∶ 10) ,respectively . The volume of 60% ethanol for elution was six times of the column volume .The recovery rate reached to 86 .76% and the purity of tota1 flavonoids was55 .27% .Conclusion The method is not only superior to the others for enriching total flavonoids but also more convenient ,which has a good application foreground .%目的:寻找一种有效富集杜仲雄花总黄酮的方法并对工艺条件进行初步摸索。方法样品溶液以聚酰胺吸附,水洗除杂。将除杂后的聚酰胺加于大孔吸附树脂柱顶部,以乙醇洗脱富集总黄酮。结果聚酰胺与 D-101树脂联合应用能很好的富集杜仲雄花总黄酮。最佳工艺条件为浸膏与聚酰胺比为1∶3,浸膏与树脂比为1∶10,以体积分数为60%乙醇洗脱,收集洗脱液6 BV 。富集物总黄酮收率可达86.76%,黄酮纯度为55.27%。结论该方法优于目前常用黄酮富集,且工艺简单,树脂再生容易,具有良好的应用前景。
