对鸡传染性法氏囊病毒(IBDV) HQ株在DF-1细胞上的适应及增殖条件进行了研究,结果表明,HQ株不适应在Vero细胞上生长,而在DF-1细胞上能够很好的适应.经培养条件优化,最终选用pH值为7.0,血清含量体积分数为2%的DMEM/F12培养基做维持液,在细胞传代后第2天接种IBDV HQ株,接毒量为1%(约0.7MOI)后36 h收毒,毒价可达8.5mL-1以上,是在CEF细胞上培养时毒价的10倍.HQ株在DF-1细胞上培养24 ~36 h上清液与全悬液毒价基本相同,维持在同一高峰.糖耗的高峰要先于病毒毒价高峰的出现,糖耗在高峰之后快速下降,当降低到接近0时病毒的毒价最高.%Study on adaption and propagation conditions of IBDV HQ strain propagated in DF-1 cells suggests that vero cells are not suitable for IBDV HQ strain to propagate but DF-1 cells are. In flask op-timization test proved that DMEM/F12 containing 2% serum, pH 7.0,1: 100 inoculation concentration of IBDV and 36 h cultivation for IBDV harvesting after its inoculation were the optimal conditions whe-IBDV was propagated on DF-1, the titers are more than 8. 5 mL-1 and 10 times as high as those propa-gated in vero cells. The titers of cell suspension and supernatant on DF-1 cell are basically the same after inoculation for 24 ~ 36 h. The peak consumption of sugar came before that of titers and decreased abruptly after the top. The titers of IBDV were the highest when the consumption of sugar was near ze-ro.
