黄芪甲苷提取物中黄芪甲苷含量测定方法比较研究

摘要

目的 比较高效液相色谱-蒸发光散射检测(high pressure liquid chromatography-evaportive light scattering detector,HPLC-ELSD)内标法和柱前衍生法测定黄芪甲苷提取物中黄芪甲苷含量的差异.方法 分别采用HPLC-ELSD内标法和柱前衍生法测定黄芪甲苷提取物中黄芪甲苷的含量,比较2种方法对测定结果的精密度、重复性、稳定性和准确度及不同批次产品测定结果的差异.结果 2种方法的测定结果差异无统计学意义,但是采用柱前衍生法测定结果的精密度、重复性、稳定性和准确度均好于HPLC-ELSD内标法.结论 采用柱前衍生法测定能更好地控制产品的质量.%Objective To compare the difference of astragaloside Ⅳ content in the astragaloside Ⅳ extract by high pressure liquid chromatography - evaportive light scattering detector( HPLC - ELSD ) internal standard method and precolumn derivatization HPLC respectively. Methods The astragaloside IV content in the astragaloside IV extract was detected by HPLC - ELSD internal standard method and precolumn derivatization HPLC respectively. The precision, repeatability, stability and accuracy of the two methods and the diversity of astragaloside IV content in different batches were compared. Results The determination results of HPLC - ELSD internal standard method and precolumn derivatization HPLC are identical statistically. But precolumn derivatization HPLC has advantages in precision, repeatability, stability and accuracy. Conclusion Precolumn derivatization HPLC can control the quality of sample better.
