肾母细胞瘤过表达基因(NOV)mRNA含量与骨肉瘤组织中细胞生长、迁移相关基因表达的相关性

         

摘要

Objective: To study the correlation of nephroblastoma overexpressed gene (NOV) mRNA levels with cell growth-and migration-related gene expression in osteosarcoma tissue.Methods: 78 patients with osteosarcoma undergoing surgical treatment in our hospital between May 2012 and May 2016 were selected, and the osteosarcoma tissue and tissue adjacent to carcinoma were collected.Fluorescence quantitative PCR method was used to detect the mRNA expression of NOV as well as pro-proliferation/anti-proliferation genes and invasion genes, and the Pearson test was used to further assess the correlation of NOV mRNA expression with the osteosarcoma cell growth and migration activity.Results: NOV mRNA expression in osteosarcoma tissue was significantly lower than that in tissue adjacent to carcinoma, FoxM1, STIM1 and Orai1 mRNA expression were significantly higher than those in tissue adjacent to carcinoma, anti-proliferation genes RanBP9, Tap73, Caspase-3, PTEN and P53 mRNA expression were significantly lower than those in tissue adjacent to carcinoma, and invasion genes EFEMP1, Notch1, Id1 and Src mRNA expression were significantly higher than those in tissue adjacent to carcinoma (P<0.05).Pearson test showed that the NOV mRNA expression in osteosarcoma tissue was negatively correlated with pro-proliferation genes FoxM1, STIM1 and Orai1 mRNA expression, positively correlated with anti-proliferation genes RanBP9, Tap73, Caspase-3, PTEN and P53 mRNA expression, and negatively correlated with invasion genes EFEMP1, Notch1, Id1 and Src mRNA expression (P<0.05).Conclusion: NOV expression is relatively low in osteosarcoma tissue, and NOV mRNA expression is negatively correlated with tumor cell proliferation and invasion activity.%目的:探讨肾母细胞瘤过表达基因(NOV)mRNA含量与骨肉瘤组织中细胞生长、迁移相关基因表达的相关性.方法:收集在本院接受手术治疗的骨肉瘤患者78例,取骨肉瘤组织及癌旁组织.经荧光定量PCR法测定其中NOV及促增殖/抗增殖基因、侵袭基因的mRNA表达量,进一步经Pearson检验评估NOV mRNA表达量与骨肉瘤细胞生长、迁移活性的相关关系.结果:骨肉瘤组织中NOV mRNA的表达量显著低于癌旁组织,FoxM1、STIM1、Orai1的mRNA表达量显著高于癌旁组织,抗增殖基因RanBP9、Tap73、Caspase-3、PTEN、P53的mRNA表达量显著低于癌旁组织,侵袭基因EFEMP1、Notch1、Id1、Src的mRNA表达量显著高于癌旁组织(P<0.05).经Pearson检验发现,骨肉瘤组织中NOV mRNA表达量与促增殖基因FoxM1、STIM1、Orai1的mRNA表达量呈负相关,与抗增殖基因RanBP9、Tap73、Caspase-3、PTEN、P53的mRNA表达量呈正相关,与侵袭基因EFEMP1、Notch1、Id1、Src的mRNA表达量呈负相关(P<0.05).结论:骨肉瘤组织中NOV呈相对低表达,且NOV mRNA表达量与肿瘤细胞增殖、侵袭活性呈负相关.
