白介素17A对口腔黏膜上皮细胞抗念珠菌黏附及β-防御素-2表达的影响

摘要

Objective:To investigate the effect of interleukin 17A (IL-17A) on the expression of β-defensin-2 (hBD-2) and the adherence of C.albicans to human oral mucosal epithelial cells (OMECs).Methods:OMECs was divided into 4 groups:a control group,an only recombinant human IL-17A (rhIL-17A) intervention group,a model group in which the cells were infected with C.albicans (1 ∶ 10 ratio),and an experimental group in which the cells were infected with C.albicans and treated with different concentrations (1,10,100 and 1,000 μg/L) of rhIL-17A.The mRNA and protein levels of hBD-2 were detected by fluorescent quantitative PCR (FQ-PCR) and enzyme-linked immuno sorbent assay (ELISA),respectively.Results:C,albicans adherence rate in the experimental group was a little lower than that in the model group (P ＞0.05).Different concentrations of rhIL-17A could up-regulate hBD-2 expression in OMECs,and 100 μtg/L was the highest (P ＜0.01).Treated with 100 μg/L rhIL-17A for 12 h,24 h and 48 h lead to markedly increased hBD-2 mRNA and protein levels in OMECs,but 48 h was the highest (P ＜0.05 or P ＜0.01).Conclusion:IL-17A could induce the hBD-2 expression in OMECs,whereas had no effect on the adherence of C.albicans to OMECs.%目的:探讨白介素17A(IL-17A)对白色念珠菌Candida albicans (C.a)感染的人口腔黏膜上皮细胞(OMECs)β-防御素-2(hBD-2)表达的影响.方法:将OMECs分为空白对照组、单独重组人IL-17A(rhIL-17A)处理组、单独C.a处理组(模型组)及C.a与rhIL-17A共同处理组(实验组).按菌:细胞=1∶10建立C.a感染的OMECs模型.分别将单独rhIL-17A处理组和实验组的OMECs按所加入的rhIL-17A浓度分为4个亚组:1μg/L组、10 μg/L组、100 μg/L组及1 000 μg/L组,共培养48 h.收集不同时间点(0h、12 h、24 h、48 h)的细胞培养上清液,计算C.a黏附率;分别采用荧光定量PCR(FQ-PCR)和酶联免疫吸附试验(ELISA)方法检测hBD-2 mRNA相对表达量及蛋白水平.结果:与模型组比较,实验组C.a黏附率有所降低,但差异无统计学意义(P＞0.05).不同浓度的rhIL-17A均能诱导OMECs表达hBD-2,且100 μg/L组hBD-2 mRNA相对表达量及蛋白水平升高最为显著(P＜0.01).采用100 μg/L rhIL-17A刺激OMECs 12 h、24 h、48 h后,hBD-2 mRNA及蛋白水平均升高,且48 h时hBD-2 mRNA及蛋白水平显著高于0h和12 h(P＜0.05或P＜0.01).结论:IL-17A能诱导OMECs表达hBD-2,但对口腔黏膜上皮抗C.a黏附无明显作用.
