熊果酸对人早幼粒白血病细胞的增殖抑制、周期阻滞及凋亡诱导作用

         

摘要

The effects of ursolic acid on proliferation,apoptosis and cell cycle on hunman promyelocytic leukemia HL-60 cells were studied using typanblue exelusion method,AO/EB stain assay and cell lab quanta.The results showed that ursolic acid inhibited cell growth in low concentration (15~20μmol/L) and lethal effect in high concentration(20~35 μmol/L) and presented in dose-dependent and time-dependent relations. The cells showed morphological characteristics of apoptosis after treated with the chemical when stained with AO/EB. The apoptosis in time and dose-dependent patterns and few necrosis cells are observed at the same time. The result of flow cytometry exhibites ursolic acid induced S phrase arrest cell in HL-60 cells. The cell cycle arrest resulted from the chemical maybe the reason that causing cell growth inhibition and apoptosis.%利用台盼蓝排染法、AO/EB双染检测法和流式细胞术检测了熊果酸对人早幼粒白血病细胞HL-60的增殖、细胞凋亡和细胞周期的作用.结果表明,低浓度(15~20 μmol/L)时药物对HL-60细胞的生长表现为抑制作用,随浓度升高(20~35 μmol/L)出现致死效应,并呈现剂量依赖和时间依赖性;AO/EB双染分析表明,药物能够诱导细胞凋亡,随着药物浓度增加和处理时间延长,细胞凋亡率上升,并且伴有少量细胞坏死;流式细胞术检测表明,在20μmol/L时,熊果酸可引起明显的HL-60细胞S期阻滞,随着浓度升高,周期阻滞作用明显加强.细胞周期阻滞可能是药物抑制细胞增殖进而引起凋亡的原因.
