龙眼体胚Obg1基因的克隆及其在龙眼体胚发生过程中的表达分析

         

摘要

OBCs are conserved small GTPases and involved in multiple cellular processes. In this experiment, three transcripts of Obgl from somatic embryos ( DLObgl) were cloned in longan (Dimocarpus longan Lour. ). Sequence analysis indicated that DLOBC1 contained typical OBC domains and showed high similarity with OBG proteins from other species, its similarity with those of Vitis vinifera , Ricinus communis and Zea mays were 84% , 84% and 82% , respectively. Real time quantitative PCR analysis indicated that transcriptional level of DLObgl changed with the development of somatic embryogenesis in longan, and the highest level occurred at heart-and torpedo-shaped embryo stages, which suggested that DLObgl be involved in formation of organs such as cotyledons.%本试验克隆了龙眼体胚Obg1基因(DLObg1)的3个转录本.序列分析表明,DLOBG1蛋白具有典型的OBG蛋白结构域,与其他植物的OBG蛋白具有较高的同源性,与葡萄、蓖麻和玉米的氨基酸序列同源性分别为84%、84%和82%.实时荧光定量PCR分析显示,DLObg1基因的转录水平随龙眼体胚的发育而变化,在心形胚和鱼雷形胚阶段的表达量最高,可能与子叶等器官的形成有关.
