液相色谱/四极杆飞行时间串联质谱筛选产β-丙氨酸甜菜碱芽胞杆菌

         

摘要

Objective To screen theBacillus-like strains producingβ-alaninebetaine.MethodsFermentation liquid samples of 32Bacillus-like strains were prepared by tryptone soy broth (TSB) medium. Extracellular metabolites of these strains were analyzed by liquid chromatography-hybrid quadrupole time-of-flight mass spectrometry (LC-QTOF MS) combined with Metlin database. ResultsAmong the selected 32 strains,Bacillus cereusstrain FJAT-16152 was the only strain whose extracellular metabolites containedβ-alaninebetaine. The content ofβ-alaninebetaine in strain FJAT-16152 was 1.47%, with the match score of 96.58%.ConclusionBacillus cereus FJAT-16152 can produceβ-alaninebetaine, which can provide theoretical basis for development and utilization of active substances fromBacillus-like strains.%目的:筛选产β-丙氨酸甜菜碱的芽胞杆菌菌株。方法用TSB液体培养基制备32个芽胞杆菌菌株的发酵液样品,采用液相色谱-四级杆飞行时间串联质谱结合 Metlin 数据库检索的方法对这些菌株的发酵液胞外成分进行分析。结果在供试的32个菌株中,只在蜡样芽胞杆菌(Bacillus cereusFJAT-16152)的胞外代谢产物中检测到β-丙氨酸甜菜碱的存在,该菌株培养液中β-丙氨酸甜菜碱的相对含量为1.47%,β-丙氨酸甜菜碱在谱库检索得分为96.58%。结论蜡样芽胞杆菌 FJAT-16152为β-丙氨酸甜菜碱高产菌株。该研究可为芽胞杆菌来源活性物质的开发与利用提供理论依据。
