脑出血大鼠脑内轴突生长抑制因子受体的表达及NEP1-40干预的实验研究

         

摘要

[目的]通过实验观察脑出血大鼠脑内轴突生长抑制因子受体(NgR)的变化规律及给予干预措施后的变化.[方法]检测脑出血大鼠脑内NGR的表达规律,同时给脑出血大鼠模型腹腔注射有干扰受体表达的干扰剂NEP1-40,考察NEP1-40对血NGR的表达抑制效果.[结果]模型成功后,脑出血大鼠出血侧的皮质及海马NGR-mRNA表达强度增加,在24 h和2 d时最高,且与模型组比较差异有显著性(P<0.01),当给予NEP1-40后,出血侧NgR-mRNA表达强度与模型组比呈现不同程度的降低,其中以24 h和2 d时间点的检测结果有差异(P<0.05).[结论]NEP1-40对脑出血大鼠出血侧NgR的表达有一定阻断能力,可为脑出血后神经功能恢复的治疗方法研究提供一个参考.%[Objective] To observe the variation of brain axonal growth inhibitory factor receptor(NGR) in rat with cerebral hemorrhage by experiment, and its change after intervention. [Methods]The variation of the expression of brain NGR in rats with cerebral hemorrhage was detected. Meanwhile, rat models with cerebral hemorrhage were intraperitoneally injected with intervention agent NEP1-40 interfering the receptor expression. The inhibitory efficacy of NEP1-40 for the expression of blood NGR was observed. [Results] After the successful establishment of the model, the expression of NGR mRNA in cortex and hippocampus at hemor-rhagic side of rats with cerebral hemorrhage increased and was up to the highest at 24h and 2d, and there was significant difference in comparison with model group ( P <0. 01). After NEP1-40 intervention, the expression of NGR mRNA decreased with different degree at hemorrhagic side in comparison with the model group, and there was significant difference in the expression level between at 24h and 2d( P <0. 05). [Conclusion] NEP1-40 can block the expression of NGR at hemorrhagic side of rats with cerebral hemorrhage so as to provide a reference for the recovery of neurological function after cerebral hemorrhage.
