单核巨噬细胞系THP-1对子宫内膜癌细胞系RL952增殖和侵袭能力的影响及其分子生物学机制

         

摘要

目的 探讨单核巨噬细胞系THP-1对子宫内膜癌细胞系RL952增殖和侵袭能力的影响及其分子生物学机制.方法 收集2015年3月至2016年5月的各类型子宫内膜病变病例180例,增殖期、分泌期、单纯增生期、复杂增生期、不典型增生和Ⅰ型子宫内膜癌病例各30例.采用免疫组织化学方法分析各类型子宫内膜病变中的巨噬细胞增殖、浸润程度.采用CCK方法检测分析单核巨噬细胞系THP-1对子宫内膜癌细胞系RL952增殖和侵袭能力的影响程度.采用Transwell法来检测分析宫内膜癌细胞系RL952对单核巨噬细胞系THP-1的招募能力.采用Western blot技术检测分析单核巨噬细胞系THP-1对子宫内膜癌细胞系RL952中的CyclinD1和MMP-2的表达水平的影响情况.结果 单核巨噬细胞系THP-1的增殖、浸润数目与子宫内膜癌细胞系RL952增殖和侵袭能力呈正相关性,差异具有统计学意义(P<0.05).子宫内膜癌细胞系RL952对单核巨噬细胞系THP-1具有招募作用.随着单核巨噬细胞系THP-1的增殖、浸润数目的增加,子宫内膜癌细胞系RL952中的CyclinD1和MMP-2表达水平随着时间增加而相应上调,差异具有统计学意义(P<0.05).结论 单核巨噬细胞系THP-1浸润为子宫内膜癌发生的机制之一.%Objective To investigate the effect of THP-1 on the proliferation and invasion of endometrial carcinoma cell line RL952 and its molecular mechanism.Methods 180 cases of endometrial lesions in our hospital from March 2015 to May 2016 were collected,including 30 cases in proliferative phase,secretory phase,simple proliferative phase,complex proliferative stage,atypical hyperplasia and endometrial carcinoma.Immunohistochemical method was used to analyze the proliferation and infiltration of macrophages in various types of endometrial lesions.CCK method was used to detect the effect of THP-1 on the proliferation and invasion of endometrial carcinoma cell line RL952.Transwell assay was used to detect the ability of THP-1 cell line RL952 to recruit monocytes.Western blot technique was used to detect the expression of CyclinD1 and MMP-2 in the endometrial carcinoma cell line RL952 by monocyte macrophage cell line THP-1.Results The proliferation and invasion of monocyte macrophage of line THP-1 were positively correlated with the proliferation and invasion of endometrial carcinoma cell of line RL952,the difference was statistically significant (P<0.05).The endometrial carcinoma cell of line RL952 has a recruitment effect on monocyte macrophage cell line THP-1.With the increase of the number of proliferating and infiltrating THP-1 in mononuclear macrophage system,the expression levels of CyclinD1 and MMP-2 in endometrial carcinoma cell line RL952 increased with the increase of time,the difference was statistically significant (P<0.05).Conclusion The infiltration of mononuclear macrophage THP-1 was one of the mechanisms of endometrial carcinoma.
