胃癌组织中RBMS3及SFRP1的表达水平与临床参数的关系及之间的相关性研究

         

摘要

目的 研究单链相互作用蛋白3(RBMS3)及分泌型卷曲相关蛋白1(SFRPl)的表达水平与胃癌的临床参数之间的关系及两者表达水平与胃癌患者预后的相关性.方法 回顾性收集新鲜人胃癌组织(23例)、对应的癌旁组织(23例),另收集172例胃癌组织及43例癌旁组织制成组织芯片.应用实时荧光定量逆转录-聚合酶链反应(qRT PCR)检测23对新鲜组织中RBMS3及SFRP1mRNA的表达水平,同时应用蛋白免疫印迹(WB)技术检测RBMS1及 SFRP1的蛋白表达水平.进一步运用免疫组化(IHC)检测172例胃癌组织及43例癌旁组织中RBMS3及SFRP1蛋白的表达水平.结果 胃癌组织、癌旁组织中RBMS3及SFRP1均有表达;RBMS3及SFRP1在胃癌组织中的表达水平明显均低于其在癌旁正常组织(P<0.05),RBMS3与SFRP1在胃癌组织中的表达水平(mRNA和蛋白)呈正相关;RBMS3在胃癌中的蛋白表达水平与胃癌的分化程度、浸润深度显著相关,SFRP1在胃癌中的蛋白表达水平与胃癌的分化程度显著相关;RBMS3与SFRP1在胃癌中的蛋白表达水平均与胃癌患者预后相关,且两者共表达水平是胃癌患者的独立预后因素.结论 RBMS3及SFRP1在胃癌组织、癌旁组织中差异表达且呈正相关;RBMS3及SFRP1共表达水平可能是胃癌患者预后的一个有价值的生物学指标.%Objective The relationship between the expression level of RBMS3 and SFRP1 and the clinical parameters of gastric cancer and the correlation between the expression level and the prognosis of gastric cancer patients were explored respectively. Methods 23 pairs of fresh human gastric carcinoma tissues and corresponding adjacent tissues were collected for quantitative real- time PCR (qRT- PCR) and Western blot(WB) test; 172 cases of gastric cancer tissues and 43 cases of adjacent tissues were used for immunohistochemical reaction. The expression levels of RBMS3 and SFRP1 mRNA and protein in 23 pairs fresh tissues were detected by qRT- PCR and WB, respectively. The expression of RBMS3 and SFRPl proteins were further detected by immunohistochemistry in 172 gastric cancer tissues and 43 adjacent tissues. Results Both RBMS3 and SFRP1 were expressed in gastric cancer and paracancerous tissues, but the expression levels of RBMS3 and SFRP1 in gastric cancer tissues were significantly lower than those in adjacent normal tissues (P <0.05). The expression levels of RBMS3 and SFRP1 in gastric cancer tissues were positively correlated. The expression levels of RBMS3 and SFRP1 in gastric carcinoma were positively correlated with the differentiation degree of gastric cancer (P < 0.05); the expression level of RBMS3 was negatively correlated with the invasion depth of gastric cancer (P <0.05). In addition, the expression levels of RBMS3 and SFRP1 were positively correlated with the prognosis of gastric cancer patients (P <0.05), and the co- expression of RBMS3 and SFRP1 was an independent prognostic factor in gastric cancer (P < 0.05 ). Conclusion The expression of RBMS3 and SFRP1 in gastric cancer tissues and adjacent tissues is positively correlated,and the co expression of RBMS3 and SFRP1 may be a valuable biological indicator for the prognosis of patients with gastric cancer.
