米诺环素对异氟醚诱导的发育期大鼠神经元毒性的保护作用

         

摘要

Objective To investigate the protective effects of minocycline on isoflurane-induced developmental neurotoxicity in primary cultured rat cortical neurons. Methods Cortical neurons of newborn SD rats were randomized into four groups (n= 10). Neurons in control group (group C) were treated with room air in 37 'C for 6 h, while isoflurane group (group A) with 1. 5% isoflurane for 6 h. Neuron in minocycline at 1 μmol/L group (group M1) and minocycline at 10 μnol/L group (group M2) were pretreated with minocycline 1 and 10 μmol/L for ten minutes, respectively, and then treated with 1.5% isoflurane Morphological changes of neurons were observed with Phase contrast microscope, apoptotic neurons were detected with terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) and neuron viability was evaluated with MTT assay. Results Neuron morphology showed swelling and deformation in group A, which was similar to group M1, while these remained virtually unchanged in group M2 and were similar to group C. Cell viability decreased while apoptotic neurons increased in group A and M1 when compared with group C and M2(P<0. 05). Conclusion Minocycline protects the primary cultured developmental rat cortical neurons from isoflurane-induced neurotoxicity.%目的 观察米诺环素对异氟醚诱导的发育期大鼠神经元毒性的保护作用.方法 取新生SD大鼠皮层,随机分为四组,每组10只.正常对照组(C组)神经元在37℃空气处理6h,异氟醚麻醉组(A组)神经元在37℃下用1.5%异氟醚处理6h,米诺环素1μmol/L组(M1组)和米诺环素10μmol/L组(M2组)神经元分别在异氟醚处理10 min前加入米诺环素1μmol/L、10μmol/L.相差显微镜下观察四组神经元形态,TUNEL法检测神经元凋亡,MTT法检测神经元活性.结果 A组神经元胞体肿胀、变形且与M1组神经元形态接近;M2组神经元形态改变不明显且与C组相近.MTT结果 显示,与C组、M2组相比,A组、M1组神经元活性显著降低(P<0.05),凋亡神经元数目显著增多(P<0.05).结论 米诺环素能减轻异氟醚诱导的发育期大鼠神经元毒性,具有保护作用.
