不同层次穴位埋线对单纯性肥胖大鼠肥胖及糖、脂代谢的影响

         

摘要

Objective:To observe the effect of catgut embedding underneath points at different layers on metabolisms of glucose and lipid in simple obesity rats.Methods:High fat diet was used to induce obesity models.The successful models were randomly divided into three groups according to different levels of embedding,which were the fat layer group,the muscle layer group and the mixed layer group.The points chosen for catgut embedding were CV12 and ST25,the intervention was once per week for four weeks.Body mass,Lees' index,fat coefficient,number and morphology of lipocyte,blood glucose,and blood fat were tested and compared among groups.Results:After catgut embedding,body mass and Lee's index of the embedding groups were lower than those of the obesity control group(P <0.01).The levels of blood glucose and blood fat in the obesity control group were significantly higher than those in the normal group (P < 0.01);the total cholesterol and triglyceride in embedding groups were decreased (P < 0.05),LDL and blood glucose were significantly decreased(P <0.01);HDL was significantly increased in the fat layer and the mixed layer embedding groups (P < 0.05).White fat were reduced in the muscle layer embedding group (P < 0.05);brown fat were decreased in all three embedding groups,in which the mixed layer embedding group decreased significantly(P <0.01);but there was no difference amog the three groups (P > 0.05).Compared to the normal group,the number and size of white fat cells and brown fat cells in the obesity control group were significantly different (P < 0.01);after treatment,the parameters of the three embedding groups were significantly different from those in the obesity control group (P <0.01).HE staining showed that fat cells increased in obese rats,the fat cells became smaller after treatment.Conclusion:Catgut embedding therapy can reduce the levels of weight,Lee's index,TC,TG,LDL-C,and blood glucose in obese rats,it can increase HDL-C level and reduce the fat content,and change the morphology of fat,so as to achieve the effect of weight loss and regulate metabolism disorders of glucose and lipid.There was no significant difference among different layers of embedding,but the mixed layer embedding has better effect on regulating glucose and lipid metabolism.%目的:观察不同层次穴位埋线对单纯性肥胖大鼠肥胖及糖、脂代谢的影响.方法:高脂饮食复制单纯性肥胖大鼠模型.将造模成功大鼠随机分为3组,按埋线的不同层次分别命名为脂肪层组、肌肉层组和混合层组.脂肪层组埋线于脂肪层,肌肉层组埋线于肌肉层,混合层组埋线在脂肪层与肌肉层交替进行.3组均取中脘穴及天枢穴行埋线治疗,频率每周1次,连续4周.治疗结束后比较各组大鼠体重、Lee's指数、脂肪系数、脂肪细胞数量与形态,并检测血脂、血糖各项指标.结果:穴位埋线治疗后,3个埋线组大鼠的体重及Lee's指数均低于肥胖对照组(均P<0.01).肥胖对照组的血脂及血糖水平较正常大鼠显著升高(P<0.01).治疗后,3个埋线组大鼠的总胆固醇及甘油三酯均下降(P<0.05),低密度脂蛋白及血糖显著降低(P<0.01),脂肪层与混合层埋线组的高密度脂蛋白显著升高(P<0.01).白色脂肪系数与棕色脂肪系数的比较,肥胖对照组均较正常对照组升高(P<0.01).治疗后,肌肉层埋线组的白色脂肪系数较模型组下降(P<0.05);3个埋线组大鼠的棕色脂肪系数均下降(P<0.05),其中混合层埋线组下降显著(P<0.01),3组之间比较无差异(P>0.05).与正常对照组比较,肥胖对照组大鼠白色脂肪细胞及棕色脂肪细胞计数与大小均有显著差异(P<0.01);治疗后,3个埋线组大鼠的上述参数与肥胖对照组比较均有显著差异(P<0.01).脂肪细胞HE染色切片可见肥胖大鼠脂肪细胞增大,治疗后脂肪细胞形态变小.结论:穴位埋线可降低肥胖模型大鼠体重、Lee's指数、血清TC/TG/LDL-C和血糖水平、升高HDL-C水平以及减少脂肪含量、改变脂肪形态,从而达到减肥和调节糖、脂质代谢紊乱的作用.3种层次穴位埋线减肥效果无显著差异,但混合层埋线对调节糖、脂代谢紊乱效果较好.
