基于显微操作的单细胞分选方法

         

摘要

In this paper, micromanipulation systems are equipped on confocal laser scanning microscope. Then we can visually select the target single cell with micropipette. Because the separation is under visible conditions,the purity of cell sorting can be up to 100%. But the sorting speed is slow(cell/2 min). This method is a way of sorting single cell, which is suitable for samples with complex composition and small sizes. It just needs microliter sample volume(2 μL at least). And it is also suitable for sorting cells with characteristic expression and special location expression, such as cells which express fluorescent proteins and cells whose exogenous genes are expressed in nucleuses and so on.%本文利用激光共聚焦显微镜成像清晰的优势,结合显微操作系统,将视野下看到的目的细胞直接用微吸管挑选出来,实现单细胞分选等功能.由于是在可视条件下进行的分选,细胞分选纯度可达100%.本方法是一种适用于样品量少、样品组成复杂、需要特性表达分选或定位分选的单细胞分选方法.
