利拉鲁肽对高脂饮食诱导的肥胖大鼠血清网膜素1及胰岛素抵抗的影响

         

摘要

目的:研究利拉鲁肽对高脂饮食诱导的肥胖大鼠血清网膜素1水平及胰岛素抵抗的影响。方法雄性Wistar大鼠40只,随机分为4组:正常对照组(NC组)、高脂饮食组(HF组)、高脂饮食+利拉鲁肽低剂量组(HF+Lira 1组)及高脂饮食+利拉鲁肽高剂量组(HF+Lira 2组)。分别饲以普通颗粒饲料及高脂饲料,12周后,HF+Lira 1及HF+Lira 2组分别给予利拉鲁肽腹腔注射4周。16周各组大鼠称取体质量,心脏取血,分别进行血糖、血脂、胰岛素、网膜素1水平测定,计算胰岛素抵抗指数。结果16周后,与NC组比较,HF组大鼠体质量、甘油三酯(tG)水平明显升高(P <0.05),网膜素1水平明显下降(P <0.05),胰岛素抵抗指数明显升高(P <0.05);总胆固醇、低密度脂蛋白胆固醇水平有所升高,高密度脂蛋白胆固醇有所下降,但无统计学差异(P >0.05)。与HF组比较,HF+Lira 1及HF+Lira 2组大鼠体质量明显下降(P <0.05),tG水平显著下降(P <0.05),网膜素1水平明显上升(P <0.05),胰岛素抵抗指数明显下降(P <0.05);HF+Lira 2组与HF+Lira1组比较,大鼠体质量明显下降(P <0.05),tG水平显著下降(P <0.05),网膜素1水平明显上升(P <0.05),胰岛素抵抗指数明显下降(P <0.05)。结论利拉鲁肽可减轻高脂饮食诱导的肥胖,改善胰岛素抵抗,可能的机制之一是与其升高血清网膜素1水平有关,且这种作用与剂量相关。%Objective to investigate the effects of liraglutide on omentin-1 and insulin resistance in high-fat diet rats. Methods Forty male Wi-star rats were randomly divided into four groups,and respectively fed with normal diet(NC group),high-fat diet(HF group),high-fat diet with small doses of liraglutide injected intraperitoneally(HF+Lira 1 group),and high-fat diet with large doses of liraglutide injected intraperitoneally (HF+Lira 2 group). After 16 weeks,the body weight,serum lipid levels,omentin-1 and insulin were measured,and then the insulin resistance in-dex were calculated. Results the body weight,the levels of tG,and the insulin resistance index in HF group were higher than NC group,omentin-1 was lower,and the differences were statistically significant(P < 0.05). tC,LDL-C were elevated,HDL-C was reduced,but there were no statisti-cally differences(P > 0.05). Compared with HF group,the body weight,the levels of tG,and the insulin resistance index in HF+Lira 1 group and HF+Lira 2 group were lower,omentin-1 was higher,the differences were statistically significant(P < 0.05). Furthermore,the effects of HF+Lira 2 group were more pronounced. Conclusion Liraglutide can restrain high-fat diet induced obesity,and improve insulin resistance through elevated the level of omentin-1 in dose-dependent manner.
