Objective To observe the effect and mechanism study of Chaihu Shugan powder on anti-arteriosclerosis in rats with metabolic syndrome(MS).Methods The rat model with MS was induced by high fat sugar and high salt diet (8 weeks), then they were fed by way of gavage with drugs for 4 weeks. The levels of weight, lipid, systolic blood pressure of arteria caudilis (SBP), fast blood glucose (FBG), superoxide dismutase (SOD), malondialdehyde (MDA), matrix metalloproteinase-9(MMP-9), tissue inhibitor of metalloproteinase-1(TIMP-1) were measured at the eighth and twelfth week, morphological changes of thoracic aorta were detected by hematoxylin eosin (HE) staining.Results Compared with the blank group,the weight, lipid, SBP, FBG, MDA, MMP-9 of rats in the model group increased significantly (P<0.05), while SOD, TIMP-1 decreased significantly (P<0.05), tatherosclerotic plaques was formed. Compared with the model group, the weight, lipid, SBP, FBG, MDA, MMP-9 of rats in Chaihu Shugan powder group decreased significantly (P<0.05),while SOD,TIMP-1 increased significantly (P<0.05), and the plaques reduced significantly (P<0.05).Conclusion Chaihu Shugan powder could delay the progression of metabolic syndrome- atherosclerosis by inhibiting the expression of MMP-9 and MDA and promoting the expression of SOD and TIMP-1.%目的 观察柴胡疏肝散对代谢综合征大鼠抗动脉粥样硬化的作用及机制研究.方法 采用高脂高糖高盐喂养方法建立大鼠代谢综合征模型(8周),造模成功后给予药物灌胃4周.实验第8周及12周检测各组大鼠的体质量、血脂、SBP、FBG、SOD、MDA、MMP-9、TIMP-1的水平,苏木素-伊红(HE)染色观察大鼠胸主动脉组织形态学变化.结果 与空白组比较,模型组在第8及12周的体质量、血脂、SBP、FBG、MMP-9、MDA的水平均明显升高(P<0.05),SOD、TIMP-1水平明显降低(P<0.05),动脉硬化形成;与模型组比较,柴胡疏肝散组第12周的体质量、血脂、SBP、FBG、MMP-9、MDA的水平明显降低(P<0.05),SOD、TIMP-1水平明显升高(P<0.05),动脉硬化减轻.结论 柴胡疏肝散可能通过抑制MMP-9、MDA的表达和促进SOD、TIMP-1的表达从而延缓代谢综合征-动脉粥样硬化的进展.
