固原鸡线粒体基因组ATPase 8基因的克隆及序列分析

         

摘要

[ Objective ] To amplify ATPase 8 genes from genomic DNA of Guyuan chicken ( white plume, ma plume and red plume), Arbor acres chicken and Roman chicken. [ Method ] Using specific primers for differential detection of chicken-derived ingredients, the ATPase 8 genes were amplified from the genomic DNA extracted from chicken blood by PCR. And the PCR products were sequenced. [ Result] The full length sequence of chicken ATPase 8 was 165 bp in the Guyuan chicken and Arbor acres chicken and 168 bp in Roman chicken. [ Conclusion ] The ATPase 8 gene of the Guyuan chicken has high homology to that of the Arbor acres chicken or Roman chicken, but it has low homology to that of quail, partridge, ostrich, goose or turkey.%[目的]扩增固原鸡(白羽、麻羽、红羽)、肉鸡品种AA鸡及蛋用品种罗曼鸡线粒体ATPase 8(ATP酶第8亚基)基因.[方法]从鸡血液中提取基因组DNA.然后,利用已经公布的鉴别检测鸡的源性成分的特异性引物,PCR扩增线粒体ATPase 8基因并测序.[结果]固原鸡不同品系和AA鸡的ATPase 8基因全序列为165 bp,罗曼鸡为168 bp.[结论]不同品系固原鸡的ATPase 8基因序列与罗曼鸡和AA肉鸡均有较高的同源性,但是与鹌鹑、鹧鸪、鸵鸟、鹅、火鸡的同源性相对较差.
